چکیده

In plants, an increasing interest for the comet assay was shown in the last decade. This versatile technique appears to be promising to detect the genotoxic effect of pollutants and to monitor the environment. However, the lack of a standardised protocol and the low throughput of the assay limit its use in plants. The aims of this paper are to identify key factors affecting comet assay performance and to improve its reliability and reproducibility. We examined the effect of varying several parameters on four different plant species: broad bean (Vicia faba), white clover (Trifolium repens), English ryegrass (Lolium perenne) and miscanthus (Miscanthus x giganteus). The influence of both internal (different nucleus isolation methods, presence or absence of filtration and lysis steps) and external (room temperature, light intensity) parameters were evaluated. Results clearly indicate that short chopping is more efficient to isolate nuclei than the standard slicing method. Filtration and lysis steps were shown to be unnecessary and thus should be skipped. Data also demonstrate that high room temperatures and light could induce DNA damage in isolated nuclei. Calibration tests with H2O2 or ethyl methanesulfonate revealed that a special attention should be paid to plant growing stage, leaf position and exposure duration.

Recommendations for conducting the in vivo alkaline Comet assay. 4th International Comet Assay Workshop.

The in vivo alkaline single cell gel electrophoresis assay, hereafter the Comet assay, can be used to investigate the genotoxicity of industrial chemicals, biocides, agrochemicals and pharmaceuticals. The major advantages of this assay include the relative ease of application to any tissue of interest, the detection of multiple classes of DNA damage and the generation of data at the level of th...

A Study of Cytogenetic Stability of Induced Pluripotent Stem Cells Using Karyotyping and Comet Assay Techniques

Background & Aims: Induced pluripotent stem cells (iPSCs) have the capability to undergo unlimited selfrenewal and differentiation into all cell types in the body. These cells are artificially derived from a nonpluripotent cell, typically human dermal fibroblasts (HDFs). The study of cytogenetic stability of these cells, in order to use iPS cells and apply studies in therapeutic applications, i...

Oecd Guideline for the Testing of Chemicals in Vivo Mammalian Alkaline Comet Assay Introduction

1. The in vivo alkaline comet (single cell gel electrophoresis) assay (hereafter called simply the comet assay) is used for the detection of DNA strand breaks in cells or nuclei isolated from multiple tissues of animals, usually rodents, that have been exposed to potentially genotoxic material(s). The comet assay has been reviewed and recommendations have been published by various expert groups...

Studies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay

Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...

Radiosensitivity and Repair Kinetics of Gamma-Irradiated Leukocytes from Sporadic Prostate Cancer Patients and Healthy Individuals Assessed by Alkaline Comet Assay

Background: Impaired DNA repair mechanism is one of the main causes of tumor genesis. Study of intrinsic radiosensitivity of cancer patients in a non-target tissue (e.g. peripheral blood) might show the extent of DNA repair deficiency of cells in affected individuals and might be used a predictor of cancer predisposition. Methods: Initial radiation-induced DNA damage (ratio of Tail DNA/Head DN...
